- This presentation describes and demonstrates the radioallergosorbent test. This objective is achieved with the aid of diagrams, graphs, views of gamma counter printouts and a step-by-step demonstration of the test. In this presentation the role of IgE in atopic disease is described first. The principles of the radioallergosorbent test are then discussed in detail. According to this presentation paper cellulose discs are activated with cyanogen bromide and then exposed to the allergen in question. The allergen becomes bound to the disc in a disc allergen IgE complex. These discs are then refrigerated until used. When a serum is to be tested, it is incubated with one of the discs. If the specific IgE is present, it will combine with the disc allergen to form a disc allergen IgE complex. I25 anti-IgE is then added to the disc allergen complex and excess anti-IgE is removed by washing with normal saline. The radiolabeled IgE can then be measured witha gamma counter. Using the series of standard discs and standard reference sera, the readings on the reference sera can be compared with the readings of the patient's sera and the amount of specific IgE in the patient's serum is calculated.
